HLA antibody screening is of vital importance to patients that require transplantation or platelet transfusion. Generally, the objectives of antibody screening in transplantation and transfusion science are threefold:
- Identifying the presence/absence of anti-HLA antibodies
- Characterising the specificity of any anti-H LA antibody, and
- Identifying the immunoglobulin class.
Products:
A) SSP Typing
SSP Typing is a PCR based technique which uses Sequence Specific Primers for DNA based Tissue Typing. Each SSP kit consists of a panel of primer mixes that each contain a specific primer pair ( i.e., the allele- and the group-specific primers) as well as a control primer pair that matches non-allelic sequences present in all samples. This control acts as an internal PCR control to verify the efficiency of the PCR amplification.
Dynal Biotech offers two formats of SSP Kit; Dynal AllSet+™ SSP and Pel-Freez SSP UniTray®.
B) RELI SSO
The Dynal RELI™ SSO typing technique utilises a generic PCR amplification with biotinylated primers that flank the specific target region of an HLA locus. This is followed by hybridisation of the resulting amplicon to an array of Sequence Specific Oligonucleotide probes immobilised on a nylon membrane. The immobilisation of these probes is referred to as a Reverse Line blot and thus gives rise to the acronym 'RELI™'.
